При многих стрессах, в том числе и при низкой температуре, в клетке происходит увеличение генерации активных форм кислорода, которые являются сигнальными молекулами и в то же время вредят клетке, повреждая ее компоненты. В свою очередь, митохондрии представляют собой одну из основных мишеней окислительного повреждения при стрессе, а кроме того, значительный источник активных форм кислорода. Митохондрии растений имеют большое количество ферментов альтернативных путей транспорта электронов, многие из которых активны при стрессе. Цель проведенного исследования заключалась в оценке влияния низкой положительной температуры и повышенной экспрессии гетерологичного гена NDB2 (альтернативная внешняя NADH-дегидрогеназа митохондрий) на генерацию активных форм кислорода, работу альтернативной дыхательной цепи в митохондриях и экспрессию стрессовых белков в условиях освещения в листьях табака Nicotiana tabacum. Установлено, что в листьях растений табака с повышенной экспрессией гена NDB2 Arabidopsis thaliana происходило снижение продукции активных форм кислорода в контрольных условиях и в условиях пониженной температуры в сравнении с растениями дикого типа. Полученные результаты свидетельствуют, что гетерологичный ген NDB2 Arabidopsis thaliana участвует в увеличении активности альтернативной электрон-транспортной цепи в митохондриях растений табака, снижает уровень генерации активных форм кислорода и влияет на содержание стрессовых белков как в контрольных условиях, так и в условиях низкотемпературного воздействия.</p></abstract><trans-abstract xml:lang="en"><p>Low temperature is an important factor limiting plant viability and productivity. Along with other stresses, low temperatures increase the generation of reactive oxygen species, which are signaling molecules that can damage cell components. As well as representing one of the main targets of oxidative damage during stress, mitochondria represent a significant source of reactive oxygen species. Plant mitochondria have a large number of enzymes providing alternative electron transport pathways, many of which are activated under stress. Our aim was to assess the effect of low positive temperatures and increased expression of the heterologous gene NDB2 (alternative external NADH dehydrogenase of mitochondria) on the generation of reactive oxygen species, which involve an alternative respiratory chain in mitochondria and the expression of stress proteins under lighting conditions in Nicotiana tabacum tobacco leaves. In the leaves of tobacco plants with increased expression of the Arabidopsis thaliana NDB2 (AtNDB2) gene, a decrease in reactive oxygen species production was observed under normal and low temperature conditions.
